SNAP-8

Also known as: SNAP-8, Acetyl Octapeptide-3

SNAP-8 (Acetyl Octapeptide-3) is a topical cosmetic peptide marketed as a needle-free alternative for softening expression lines by reducing muscle contraction at the surface. It is an elongated analog of the Argireline hexapeptide.

Mechanism of Action

SNAP-8 is Acetyl Octapeptide-3, an eight-amino-acid peptide derived from the N-terminus of SNAP-25. It is designed to compete with native SNAP-25 within the SNARE protein complex required for neurotransmitter vesicle docking and fusion at the neuromuscular junction. By interfering with SNARE complex assembly, it is theorized to reduce acetylcholine release and thereby attenuate the strength of facial muscle contractions that produce dynamic expression lines. Applied topically, the effect is local and reversible, in contrast to the enzymatic cleavage produced by botulinum toxin, and it is frequently formulated alongside Argireline (Acetyl Hexapeptide-3/8).

Reported Benefits

  • May soften the appearance of dynamic expression lines
  • Topical, needle-free approach to wrinkle reduction
  • Targets the SNARE complex to reduce muscle contraction signaling
  • Often combined with Argireline for additive effect
  • Generally well tolerated in cosmetic formulations

Potential Side Effects

  • Possible mild local irritation or redness
  • Rare contact sensitization
  • Transient tightness or dryness at the application site

Common Dosing

Research-use only and not medical advice; in cosmetic research Acetyl Octapeptide-3 is commonly formulated at roughly 3-10 percent of a peptide-solution concentrate in topical products.

Contraindications

  • Known hypersensitivity to the peptide or formulation excipients
  • Application to broken or irritated skin

Botox is injected and enzymatically cleaves SNARE proteins to block muscle contraction, while SNAP-8 is a topical peptide that competitively interferes with SNARE complex assembly. The topical effect is milder, local, and reversible.

SNAP-8 is an elongated octapeptide analog of the Argireline hexapeptide. Both target the SNARE complex, and they are often used together in cosmetic formulations.
