CJC-1295 without DAC is a modified GHRH(1-29) analog that binds the pituitary GHRH receptor to amplify the amplitude of natural growth-hormone pulses. GHRP-2 is a synthetic ghrelin-mimetic that activates the growth-hormone-secretagogue receptor (GHS-R1a), triggering GH release through a distinct pathway and partially suppressing somatostatin tone. Combining a GHRH analog with a GHRP produces a synergistic, greater-than-additive GH pulse because the two receptors act on complementary signaling cascades. The no-DAC form has a short half-life that preserves pulsatile, physiologic GH and downstream IGF-1 release.
