CJC-1295 without DAC is a modified GHRH(1-29) analog that binds the pituitary GHRH receptor and increases the amplitude of endogenous growth-hormone pulses. GHRP-6 is a synthetic hexapeptide ghrelin mimetic that activates the GHS-R1a receptor to trigger GH release and reduce somatostatin inhibition, and it is the most potent of the GHRPs for stimulating appetite via central ghrelin signaling. Pairing a GHRH analog with GHRP-6 yields a synergistic GH pulse because the two receptor systems reinforce one another. The short half-life of the no-DAC form keeps GH and IGF-1 release pulsatile and physiologic.
