Research framing extends the amylin-add-on logic of CagriSema to a triple agonist: GLP-3 R acts on GLP-1, GIP, and glucagon receptors, while cagrilintide (a long-acting amylin analog) adds a hindbrain fullness pathway none of those receptors cover. The evidence for this specific pairing is thin: a single rodent study (obese male rats) reported greater weight loss with the combination than with either agent alone, and there is no published human trial. Both components are investigational, and stacking two potent appetite suppressors compounds GI side effects such as nausea and vomiting. Once-weekly subcutaneous dosing is typical of each component in trials.
