Somapacitan

Also known as: Sogroya, Somapacitan-beco, NNC0195-0092

A once-weekly long-acting growth hormone, FDA-approved as Sogroya for adult (2020) and pediatric (2023) GH deficiency. Prescription medication.

Mechanism of Action

Somapacitan is human growth hormone with a single amino-acid substitution to which a fatty-acid side chain is attached. The side chain binds reversibly to circulating albumin, slowing clearance and extending the half-life so one injection a week is enough. It acts at the growth hormone receptor exactly as native GH does, raising IGF-1 and producing GH's growth and metabolic effects.

Reported Benefits

  • Once-weekly replacement for adults with GH deficiency
  • Height velocity non-inferior to daily somatropin in children with GH deficiency (REAL4)
  • Label expanded in 2026 to children born SGA, with idiopathic short stature or with Noonan syndrome

Potential Side Effects

  • Injection-site reactions
  • Headache, nasopharyngitis, arthralgia
  • Peripheral edema
  • Reduced insulin sensitivity and higher blood glucose
  • Class GH warnings: intracranial hypertension, slipped capital femoral epiphysis, scoliosis progression

Common Dosing

Labeled dosing is once weekly by subcutaneous injection: weight-based in children (0.16 mg/kg per week for GH deficiency) and a fixed starting dose in adults (1.5 mg weekly, higher in women on oral estrogen) that is titrated to IGF-1 and tolerability. Dosing is set and supervised by the prescribing clinician.

Yes. Sogroya was FDA-approved for adult growth hormone deficiency in August 2020 and for pediatric growth hormone deficiency in April 2023. In February 2026 the FDA added children born small for gestational age, idiopathic short stature and Noonan syndrome.

It is the same hormone modified to bind albumin, so it lasts long enough for weekly rather than daily injections. In phase 3 trials its effect on growth and IGF-1 was comparable to daily somatropin.
