Dulaglutide

Also known as: Trulicity, LY2189265

Once-weekly GLP-1 receptor agonist, FDA-approved in 2014 as Trulicity for type 2 diabetes and cardiovascular risk reduction. Prescription medication.

Mechanism of Action

Dulaglutide consists of two GLP-1 analog peptides fused to a modified human IgG4 Fc fragment. The large fusion protein resists DPP-4 breakdown and is cleared slowly, allowing once-weekly dosing. It activates the GLP-1 receptor to increase glucose-dependent insulin secretion, suppress glucagon, slow gastric emptying and reduce appetite.

Yes. The FDA approved it as Trulicity in 2014 for type 2 diabetes, added a cardiovascular risk-reduction indication in 2020 based on REWIND, and extended use to children 10 and older in 2022.

No. It is approved for type 2 diabetes; weight loss is a side benefit and is smaller than with semaglutide or tirzepatide.
