Triptorelin is a decapeptide GnRH agonist that initially stimulates pituitary release of LH and FSH, producing a transient hormone flare. With continuous receptor occupancy the gonadotrophs become desensitized and downregulated, sharply reducing LH, FSH, and downstream testosterone or estradiol. This reversible medical castration underlies its use in hormone-dependent cancers and conditions. Depot formulations provide sustained receptor stimulation over weeks to months.
