N-Acetyl Semax Amidate is an analog of the ACTH(4-10) fragment Semax in which N-acetylation and C-terminal amidation enhance stability against peptidases. It strongly upregulates brain-derived neurotrophic factor (BDNF) and its receptor TrkB, supporting neuroplasticity, neuronal survival, and synaptic function. Semax analogs also modulate the dopaminergic and serotonergic systems and inhibit enkephalin-degrading enzymes, contributing to improved focus, motivation, and stress resilience. These combined neurotrophic and neuromodulatory actions underlie its investigated nootropic and neuroprotective effects, including in ischemic models.
