ARA-290 is a short peptide modeled on the helix-B region of erythropoietin (EPO) that selectively activates the innate repair receptor, a heteromer of the EPO receptor and the beta-common (CD131) receptor. Unlike full-length EPO, it does not bind the homodimeric EPO receptor, so it produces tissue-protective and anti-inflammatory signaling without stimulating erythropoiesis. Activation of this pathway dampens pro-inflammatory cytokine release, promotes survival of neurons and small nerve fibers, and supports tissue repair, which is why it is investigated in neuropathic pain and sarcoidosis.
